Résumé scientifique

The tick-borne Crimean-Congo hemorrhagic fever virus (CCHFV) causes severe and often fatal hemorrhagic disease for which no approved countermeasures are available. Herein, we identify the phosphatidylserine receptors (PSRs) AXL, TIM-1, and TIM-4 as critical entry host factors for CCHFV in cell lines and liver-derived primary cell cultures. Small-molecule and antibody-based PSR inhibitors greatly reduced viral infection in cell culture. Germline knockout of individual PSR genes afforded partial protection against CCHFV infection, disease, and death in a murine model of lethal viral challenge. Mice engineered to lack two PSRs were completely protected from challenge, with viremia and viral replication, pathology, and inflammation in the liver and spleen reduced to baseline. Therefore, PSRs are collectively indispensable for CCHFV infection and pathogenesis. We conclude that PSRs are not just cell entry factors but bona fide determinants of infection, dissemination, and disease by CCHFV and legitimate targets for the development of new antiviral countermeasures.

    Résumé scientifique

Crimean-Congo haemorrhagic fever (CCHF) is a tick-borne zoonotic disease caused by the Crimean-Congo hemorrhagic fever virus (CCHFV). Ticks belonging to the genus \textit{Hyalomma} are the main vectors and reservoir for the virus. It is maintained in nature in an endemic vertebrate-tick-vertebrate cycle. CCHFV is prevalent in wide geographical areas including Asia, Africa, South-Eastern Europe and the Middle East. Over the last decade, several outbreaks of CCHFV have been observed in Europe, mainly in Mediterranean countries. Due to the high case/fatality ratio of CCHFV in human sometimes, it is of great importance for public health. Climate change and the invasion of CCHFV vectors in Centra…

Mathematical Modeling of CRISPR-CAS system effects on biofilm formation
Qasim Ali, Lindi Wahl · 2016 · Populations and Evolution · cité 6 fois · Résumé IA disponible

Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR), linked with CRISPR associated (CAS) genes, play a profound role in the interactions between phage and their bacterial hosts. It is now well understood that CRISPR-CAS systems can confer adaptive immunity against bacteriophage infections. However, the possibility of failure of CRISPR immunity may lead to a productive infection by the phage (cell lysis) or lysogeny. Recently, CRISPR-CAS genes have been implicated in changes to group behaviour, including biofilm formation, of the bacterium Pseudomonas aeruginosa when lysogenized. For lysogens with a CRISPR system, another recent experimental study suggests that bacteriophage re…

Killing of targets by effector CD8$^+$T cells in the mouse spleen follows the law of mass action

It has been difficult to measure efficacy of T cell-based vaccines and to correlate efficacy of CD8$^+$T cell responses with protection against viral infections. In part, this difficulty is due to our poor understanding of the in vivo efficacy of CD8$^+$T cells. Using a recently developed experimental method of in vivo cytotoxicity we investigated quantitative aspects of killing of peptide-pulsed targets by effector and memory CD8$^+$T cells, specific to three epitopes of lymphocytic choriomeningitis virus (LCMV), in the mouse spleen. By analyzing data on killing of targets with varying number of epitope-specific effector and memory CD8$^+$T cells, we find that killing of targets by effector…

Infection by many viruses begins with fusion of viral and cellular lipid membranes, followed by entry of viral contents into the target cell and ultimately, after many biochemical steps, integration of viral DNA into that of the host cell. The early steps of membrane fusion and viral capsid entry are mediated by adsorption to the cell surface, and receptor and coreceptor binding. HIV-1 specifically targets CD4+ helper T-cells of the human immune system and binds to the receptor CD4 and coreceptor CCR5 before fusion is initiated. Previous experiments have been performed using a cell line (293-Affinofile) in which the expression of CD4 and CCR5 concentration were independently controlled. Afte…

The role of parasite-produced dopamine in Toxoplasma gondii-altered host behaviour

Abstract Certain parasites appear to manipulate their host’s behaviour for their own selective benefit. The mechanisms behind such behavioural alterations in vertebrate hosts remain elusive, despite their profound implications, particularly for those parasites with a broad host range. Toxoplasma gondii , the causative agent of the globally important toxoplasmosis, is a canonical example for parasite-altered behaviour in its rodent intermediate host. Such behavioural alterations, from subtle to severe, are also displayed across a broad spectrum of infected secondary hosts, including humans. Dopamine dysregulation has been proposed as one potential mechanistic explanation. Furthermore, T. gond…

Dengue is the most prevalent arthropod-borne viral disease. Clinical symptoms of dengue virus (DENV) infection range from classical mild dengue fever to severe, life-threatening dengue shock syndrome. However, most DENV infections cause few or no symptoms. Asymptomatic DENV-infected patients provide a unique opportunity to decipher the host immune responses leading to virus elimination without negative impact on t v 'health. We used an integrated approach of transcriptional profiling and immunological analysis comparing a Cambodian population of strictly asymptomatic viremic individuals with clinical dengue patients. Whereas inflammatory pathways and innate immune responses were similar betw…

A parasitoid overcomes collective defense by silencing host communication
