Influenza viruses are constantly evolving, allowing them to evade the host's immune system. This evolution is driven by changes in the virus's antigenic phenotype which can be measured using the hemagglutination inhibition (HI) assay. In this study, the authors build upon previous approaches to antigenic cartography and develop a model that combines both genetic and antigenic evolution of influenza viruses. Using data from different influenza lineages (A/H3N2, A/H1N1, B/Victoria, and B/Yamagata), they analyze patterns of antigenic drift across viral lineages. They find that A/H3N2 evolves at a faster rate and in a more punctuated fashion compared to other influenza lineages. Furthermore, it is shown that year-to-year antigenic drift plays a significant role in driving incidence patterns within each influenza lineage. This suggests that understanding antigenic drift is essential for predicting and managing influenza outbreaks. The integration of molecular and antigenic evolution in this model opens up new possibilities for studying the dynamics of not only influenza but also other pathogens with variable antigens. By providing an extensive understanding of how viruses evolve both genetically and antigenically, this research paves the way for future advancements in investigating and combating infectious diseases.
